Abstract

Nitric oxide (NO) is known for its crucial role in the regulation of endothelium-dependent vasorelaxation. However, it is now recognized that NO mediates a variety of physiologic processes, including the immune, nervous, and pulmonary systems. Since exercise causes changes in vasorelaxation and blood flow, the present study examined whether a month exercise could affect gene expression of endothelial nitric oxide synthase (ecNOS) in the rat kidney. To estimate the gene expression of ecNOS at molecular levels, we measured the amounts of protein and mRNA with Western blotting and reverse transcription- polymerase chain reaction (RT-PCR), respectively, Protein and mRNA levels of ecNOS were increased by exercise compared to sedentary controls, and the extent of increases was parallel to the duration of exercise. These results indicate that ecNOS gene expression is induced by exercise, and that the increased ecNOS may help in part to explain the molecular basis for beneficial effects of physical exercise on regulation of blood flow and renal circulation.
